The couple, who play Debbie Dingle and David Metcalfe in the soap, have a three-year-old son together, called Buster.
A spokeswoman for the show confirmed the pair had decided to end their relationship.
She said: "Charley Webb and Matthew Wolfenden have confirmed that they have separated.
"They will remain friends and are focusing on their little boy."
Yesterday (7 August) Wolfenden added to the message via his Twitter account writing: "Thank you so much for all your lovely messages. Unfortunately these things happen, and it's very sad. Buster is our main priority always. X."
Webb, 25, and Wolfenden, 33, dated for five years. In 2009, they announced their engagement.
Last year, the actor was quizzed on his upcoming nuptials with his fiancée. At the time, he explained the pair were to busy to plan their wedding.
He added: "We don't disagree about what we want to do for our wedding. But up 'til now, we haven't had a minute to plan it."
