The 30-year-old and her husband, John Krasinski, are said to be "delighted" with the news.
A representative for the actress confirmed the news to Us Weekly magazine, adding that she is due to give birth in a few months.
Blunt, who is originally from Britain, has been with 33-year-old Krasinski since November 2008. They tied the knot in July 2010 in Como, Italy.
Last year, the actress admitted in an interview with Manhattan magazine that the couple had been discussing starting a family.
She said: "We want to have children. I just don't know when yet. I'm just not sure about the timing of all of that. We're both from big families, so I think it's something we want."
