The couple have been married for more than 10 years and have two children together.
In a statement, which was released on Paltrow's website Goop on Tuesday, the pair said they had decided to take the "sad" decision after trying to save their marriage for more than a year.
The statement, entitled Conscious Uncoupling, read: "It is with hearts full of sadness that we have decided to separate. We have been working hard for well over a year, some of it together, some of it separated, to see what might have been possible between us, and we have come to the conclusion that while we love each other very much we will remain separate.
"We are, however, and always will be a family, and in many ways we are closer than we have ever been. We are parents first and foremost, to two incredibly wonderful children and we ask for their and our space and privacy to be respected at this difficult time.
"We have always conducted our relationship privately, and we hope that as we consciously uncouple and coparent, we will be able to continue in the same manner."
Within minutes of the message being posted, Goop crashed due to the volume of people trying to visit the site.
Paltrow, 41, and Martin, 37, married in a secret ceremony in December 2003, just days after they confirmed the actress was expecting their first child. The couple had already been together for a year, after reportedly meeting backstage at a Coldplay concert.
They have two children together, a nine-year-old daughter named Apple, and a son, Moses, aged seven.
