News of the split was first reported by The Sun newspaper, who claim the pair have agreed to a trial separation. It is thought Stone has moved out of the family's London home, and has taken the couple's son and their pet dog with her.
A source is quoted by the publication as saying: "David has been putting on a brave face. But people close to him are very worried because they know he's devastated."
They added that the comedian is "determined" to win back his wife.
A representative for the 43-year-old has refused to comment on the claims.
Walliams, and Stone, 31, started dating in 2008 and announced their engagement in January 2010. That May, the couple tied the knot at London's Claridge's Hotel. Two years later, in December 2012, they confirmed they were expecting their first child together. Their son, Alfred, was born in May 2013.
